In this study , on the base of Transgenic Hybrid Poplar 741 which Zheng Junbao and Tian Yingchuan work on , successful infected A. rhizogenes strain15834.Compared with control Transgenic Hybrid Poplar 741 and the transgenic plantlet of the A. rhizogenes on 741 Chinese white poplar. Study on polygenes transformed and polygenes expression.Transformed multiple genes plantletlet was confirmed by the shape characteristic, PCR analysis(rolD and MAS genes, NptII and BtCry I Ac genes). Indicate polygenes have already been combined into in the regeneration plantlet.1. To polygenes transformed subclones department test in the tissue culture who take root, root length, root quantity, stem high and phenotype make a variation research of carrying on. Result indicate poplar 741 containing poly-transgenes compared with control, The leaf which transplant is in MSo medium have a characteristic of the spontaneous root, from the shape characteristic the transgenic plantletlet take place leaf wrinkle,the stem growth is shorter, raise group bank up rate of taking root of shoot with earth obviously, lateral root developed quickly and strong,etc. 98% about transplant subclone root quantity of department increase constrasting obviously. Lateral root developed, take shape and form a cluster and take root,the number is countless. Root length a few subclones department higher than constrast obviously, have some subclones department and constrast plantlet to be similar only. Have some subclones root of department long to shorter than constrast obviously.After the translet of multiple genes , Stem high compared with constrast have a remarkable difference among leaf shape , leaf area , petiole length , section, majority show as leaf wrinkle, long/wide increase , leaf area reduce , petiole length shorten , high characteristic of reducing etc. of growing etc. leave.2. Determined the endogenous hormone of some plantlet leaf of subclones through HPLC method, IAA content is higher than constrast plantlets notably in all transgenic plantlet leaf. By applying enzyme-linked immunosorbent assay method, the content of Bt(Bacillus thuringiensis Berliner) toxic proteins in some poly-transgenic plantlet leaf and root of subclones was tested.The results showed that Bt toxin proteins was detected in all the test organs of the transgenic, its contents varied significantly among leaf androot ,leaf 's Bt toxic proteins expressed was higher than root.Most of the subclone Bt toxic proteins content in the leaf and root is lower than Transgenic Hybrid Poplar 741 Carrying Two Insect-resistant Genes,but higher than A., rhizogenes.Indicated that transform from strainl5834,can lower the content of the Bt toxic Proteins.The transformed strainl5834 heavily infection the content of the Bt toxic proteins,the higher and lower even infect on every indie of plant,especially the growth of the plant.The endogenous hormone is a sample,in contrary,infect the growth of the plant,even infect the Bt toxic protein persistence and degradation. |
