SSR Fingerprinting Establishment Of Different Cultivars Of Peach, Plum, Apricot And Walnut

Peach (Prunus persica (L.) Batch.);Plum (Prunus salicina L.);Apricot (Armeniaca vulgaris L.);walnut (J.regia L.) had a long life and the main breeding method was clone. Species exchang existed in different area which leaded to different species had a same name and different name belonged to the same species. So it was important to safeguard the rights and interests of the breeding worker. In this article, 18 primers were selected to differentiate 20 peach cultivars, 20 plum cultivars, 21 apricot cultivars by SSR.and 10 primers were selected to differentiate 21 walnut cultivars. At the same time we contructed the fingerprints of peach, plum, apricot and walnut, similarity coefficient was analysed. The main results were as follows:Selecting out 18 pairs of polymorphic primers from SSR primers of peach, 20 peach cultivars, 20 plum cultivars, 21 apricot cultivars were analysed by SSR, 216 bands of peach were amplified, per pair of primer could amplified 12 bands, among of them , polymorphic bands were 210 and the percentage of polymorphic band was 97.22%. Plum cultivars were amplified by 16 pairs of primers, 189 bands were amplified, per pair of primer could amplified 11 bands, polymorphic bands were 183 and the percentage of polymorphic band was 96.83%. Apicot cultivars were amplified by 15 pairs of primers, 140 bands were amplified, per pair of primer could amplified 9.3 bands, polymorphic bands were 139 and the percentage of polymorphic band was 96.43%. The result of researching means that the genetic diversity in the intraspecies of peach, plum, apricot was high, heredity variation of peach is higher than plum and apricot.Selecting out 10 pairs of polymorphic primers from SSR primers for walnut. 128 bands were amplified, per pair of primer could amplified 12 bands, polymorphic bands were 114 and the percentage of polymorphic band was 89.06%. This means genetic diversity of walnuts in intraspecies was high.In 18 pairs of primers, apicot could be identified from peach and plum with primer UDP96-013 or UDP98-414, peach could be identified from apricot and plum with primer P08, plum could be identified from peach and apicot with primer BPPCT 026. 6 peach cultivars: Xinchuanzhongdao, Xuetao, Hanlumi, Qiuhong, BixiaPantao, Zaohuang;4 plum cultivars: Moerteni, Meilili, Nvshen, Weixin;3 apricot cultivars: Jidanxing, Jinxidahongxing, Hongmeixing had specific bands. These cultivars could be identified by specific primers. The other cultivars could be identified through ErQi Classification. P04was the most efficient primer in identifying peach, plum, apricot cultivars in the intraspecies, 14 peach cultivars, 16 plum cultivars, 19 apricot cultivars were identified.In 10 pairs of polymorphic primers, Fengyang and walnut cultivars could be identified by primer WGA4, WGA11, WGA17. Aimige, Anbian4, Dai2 had specific bands. Aimige could be identified from other walnut cultivars with primer WGA25, Anbian 4 could be identified from the other walnut cultivars with primer WGA6, Dai2 could be identified from the other walnut cultivars with primer WGA11. The other cultivars could be identified through ErQi classification.The number of amplified bands of WGA32 was 24 which was more than other primers and all the bands were polymorphic bands. The WGA32 was the most efficient primer in identification walnut cultivars.The fingerprinting of 61 germplasm of peach, plum, apricot materials were established based on all the SSR bands using 18 primers. These germplasm could be separated into 3 groups by UPGMA cluster analysis method. Their genetic diversity was discussed. The fingerprinting of 22 germplasm of walnut and Fengyang materials were established based on all the SSR bands using 10 primers by Jacard method. Their genetic diversity was analyzed. It accounted the complexity and similarity of genetic background of the materials, and provided theory foundation for species identification and essentialism protection.
