Studies On Drugs Of Jujube Witches' Broom Filtration In Tissue Culture Condition

Jujube witches' broom (JWB) cause by phytoplasma, is becoming the most destructive infectious disease of Chinese jujube and transmitted almost every jujube planting area in the world, it has brought much economic damage to our country. Hormone, element, antibiotics, Chinese traditional medicine, temperature and pH value were studied in vitro culture condition to find JWB cure measures, and cure factors were analyzed in this experiment, efficient drugs were found. The main results were as follows:1. Observed a great deal of JWB plantlet, we found that, small leaves, short burl distance, thin stem, more axillary bud, stipule thorn became leaves bigger, is the JWB representative symptom;JWB symptom standard were defined into four grade, the relation between disease symptom and state of illness become definite.2. At the point of auxins level is low and hormone proportion was maladjusted in JWB plantlet body, different kind of hormone and different proportion were applied in culture medium. It was found that the plantlet were only cure into the 2 or 1 disease symptom standard by hormone, treatment of auxins united with cytokinins is prefer to auxins treated lonely, the best hormone treatment is NAAl.Omg-L^-1 +KT1.0mg-L^-1, which could transform JWB plantlet into 1 disease symptom standard at 60d.3. Kinds of element were applied in culture medium, it was found that 1.48g·L^-1 MgSO₄·7H₂O treatment could cure the JWB disease provisionally after 70 days treated, 0.1668g-L^-1 MnSO₄·H₂O treatment could only cure plantlet to 2 disease symptom standard, but its cured rate was higher. FeSO₄·7H₂O CaCl₂·2H₂O could also cure plantlet to 2 disease symptom standard, but their cured rate were low and effect were instable.4. The result of JWB drugs filtration show that, macrolide drugs were the best treatment, 100μg-mT^-1 roxithromycin could cure disease plantlet thoroughly at 60d, and its cure speed and cured rate were the highest, restrain effect were the lightest, its cure effect was superior to tetracycline and oxytetracyline, and its future of application in field was bright. Azithromycin could also cure plantlet thoroughly, but it poison plantlet seriously, survival rate and cured rate were low. Quinolones drugs' cure effect was light and restraint effect was conspicuous. Bio-repellent 0116 had no cure effect on JWB plantlet.5. Low temperature and high pH value could cure the plantlet more or less, transform the plantlet to 2 disease symptom standard, but they restrain the growth speed conspicuously, survival and cured rate were low. These treatments could only qua adassistant factors in JWB cure technique.
