Anaplasmosis is a hemoparasite and tick-borne disease of cattle, sheep and other ruminants caused by Anaplasma marginale. Anaplasmosis can mainly cause the death of cattle ,sheep ,deer and other ruminants. The mortality of cattle can be over 80%. Now the OIE and the Ministry of China Agriculture takes Anaplasmosis as the more important diseases in the animal quarantine.Now there are many serological methods to detect Anaplasmosis :CF, CAT, c-ELISA, dot-ELISA, IFAT, DNAprobes, PCR and so on. But there are so many shorts of those means such as fussy operationhigh technology leve, special instrument, expensive, long time and so on which are not good to immunity inspect or diagnoses. So those shorts limit their use in grass roots. Then LAT are already extensively used in disease diagnoses because of their advantage: fast, convenience , high sensitivity and specificity.The testes cattle was inoculated with Anaplasma marginale which was preserved in the liquid nitrogen. When parasitemia reached the high peak, blood sample was collected anticoagulatively , then these blood was injected into another test cattle by vein. When parasitemia reached the high peak, the blood was collected. Then erythrocytes were broken by 0.85% NH4CL, then centrifugation, upper solution was pulled, and down part, Anaplasma marginale was broken by ultrasound. The soluble antigen can be used in the latex agglutination test.Using the soluble antigen of Anaplasma marginale to sensitizing the latex by chemical cross-link, a latex agglutination test(LAT) was developed for detection of serum antibody against A.marginale The sensitized latex showed no reaction with the antibodies against Theileria sergenti, Babesia bigemina, Chlamydiae, Eperythrozoon diseases; and obvious agglutination with standard positive serum, immunitized bovine serum and samples of serum from epidemic area. The result indicated that the LAT is a simple rapid sensitive specific inexpensive method, and has wide application in clinical practice, especially to rapid dignosis of Amarginale disease in the field.
