The immunohistochemical method of SABC(Streptavidin-Biotin-Peroxidase Complex)was used to study the distributed regularity, target organs, target cells of Pseudorabies virus (PRV) , the relationship between PRV existent position and pathological changes in piglet tissues which had been infected pseudorabies virus-EA artificially. The tissue paraffin slides of piglets were stained by SABC, and observed by optical microscope. And the negative tissues were the tissue of the healthy piglets which had not been infected PRV that was defected by LA.The IgG of rabbit anti-PRV was the primary antibody in this trial. At first, the PRV as antigen was purified by overspeed centrifugal method. Four rabbits were inoculated in the light of the general method. Antisera were collected when the titer was matched with the experiment. The IgG (i.e. the primary antibody) of the antisera was purified by polyglucosan G-200, and it was specificity which was identified by double agar diffusion test. The working concentration of IgG was 1:600.SABC staining and observing shows that there were positive cells in lung, cerebrum, cerebellum, spinal cord, spinal ganglion, lymph node, tonsil, thymus, spleen, liver, kidney, adrenal gland, gastro and intestine et al when the PRV intruded the body of piglet, especially in lung, nervous tissue and lymphoid organizations. The pseudorabies virus strain invaded epithelial cell, neurocyte and lymphocyte, and existed in cytoplasm and karyon of infected cells widely,mostly in cytoplasm. There was not positive cell in all the control tissues.H.E. staining and observing shows that the most pathological changes were interstitial pneumonia and hemorrhagic pneumonia, typical nonsuppurative encephalitis, and neurogangliitis et al. The lymphoid tissue was necrosis, and the amount of lymphocytes was reduced. The pathological changes of the positive cells, especially the epithelial cells were swell, denaturization and hyperplasy to the extent.The compare of the staining of SABC and H.E. shows that the pathological changes were clearly correlated with the position of the PRV existent position. The lung, nervous tissues and lymphoid tissues of piglet were damaged by PRV-Ea strain directly. |
