| Three Experiment were conducted to determine the effect of EGF and IGF-1 on in vitro maturation of goat oocytes. Oocytes were cultured in Oocytes Maturation medium with EGF and IGF-1 alone or combinedly at different concentration. The results in experiment 1 showed that there was no significant different between control group and the group with EGF(10μg/L﹑20μg/L﹑30μg/L) in maturation rate. In experiment II the maturation rate of goat oocytes was improved significantly in the groups with EGF(20μg/L﹑30μg/L) (P<0.05),but not in the group with 10μg/LEGF. In experiment III the maturation rate in the group containing 10μg/LEGF and 20μg/LIGF-1 was significantly higher than 20μg/L IGF-1 and 10μg/LEGF groups.The results obtained in this study indicate that EGF and IGF-1 have coeffects on in vitro maturation of goat oocytes. In vitro maturation oocytes were treated with CB or without CB treated. After parthenogenetic oocytes were cultured in vitro in CR1aa (groupâ… )or SOFaa(group Ⅱ) medium supplement with 10 percent of FBS or EGS. The results of this study showed that in experimentâ… ã€€there was no significant different between CB treated and without CB treated group in the ratio of cleavage and blastocyst of parthenogenetic goat oocytes; the blastocyst rate of embryos  cultured in CR1aa supplemented with 10 percent of EGS was higher significantly than supplemented with 10 percent of FBS. In experimentⅡ there was no significant different between CB treated and without CB treated group in cleavage and blastocyst rate of parthenogenetic goat oocytes; the blastocyst rate of parthenogenetic goat oocytes cultured in SOFaa supplemented with 10 percent of EGS was higher significantly than supplemented with 10 percent of FBS. It showed that CB  was no significant effect on the development of goat parthenogenetic oocytes. Culture medium supplemented with 10 percent of EGS has higher significantly effect than FBS. SOFaa was better than CR1aa in culturing goat parthenogenetic oocytes. Nuclear transfer was carried out with goat fetal fibroblasts cells cumulus granulosa and human fetal fibroblasts cells by means of intracytoplast injection and electro-fusion respectively. The results of intracytoplast injection showed that the development capacity of cloned embryos from goat fetal fibroblasts cells and cumulus granulosa cells were higher significantly than human fetal fibroblasts cells. Electro-fusion result of goat fetal fibroblasts cells was higher significantly than human fetal fibroblasts cells. Conclusion: human fetal fibroblasts cells had development capacity in goat denucleared oocytes but it was lower significantly than goat fetal fibroblasts cells and  cumulus granulosa cells. |
