Barley seeds were treated by different high hydrostatic pressure or were kept for different time. As comparison to control, both germination rate and seedling rate decreased considerably with pressure or treated time increasing . The above results showed that high hydrostatic pressure could affect the seeds germination ,seedling growth.. Plant growth was inhibited in treated materials during the early growth stage of seedling. The genetic variation of the different high hydrostatic pressure and the different time treated material were investigated by ISSR and RAPD analysis, 20 ISSR and 19 RAPD primers generated 937 and 600 bands, of which 382(40.76%) and 266 (44.33%) were polymorphic respectively .For T2 and P3,173 and 187 bands were generated with 66.21% and 68.77% being polymorphic. ISSR analysis indicated that the mean absolute distance coefficient between the original breed and the T2 is 0.6047, while that between the original breed and the P3 is 0.5636. RAPD analysis demonstrated that the mean absolute distance coefficient between the original breed and the T2 is 0.6168, while that between the original breed and the P3 is 0.5949. Some specific amplified DNA fragments was obtained from genomic DNA of high-pressure induced mutagenized barley using ISSR and RAPD technique.After cloning this fragment into p GEM-T easy vector and sequencing it, we found that ISSR and RAPD analysis of these two mutant and the original breeds revealed that there are significantly polymorphism between the two mutants and the original breed, each with 58.59% and 49.93%. DNA methylation is the main cause of transcription algensilencing. ISSR analysis indicated that there existed significant difference in DNA methylation status between the original breed and the high hydrostatic pressure treated.
