We have obtained a gene involved in salt-resistance via anther cultivating and EMS induced mutations (salt-resistance mutation RH8706-49 and salt-sensitivity mutation H8706-34 ) from one seed in our lab, which was named glycogen synthase kinase 1 in Triticum asetium(TaGSftl). As a new found gene , TaGSkl had been accepted (AF525086) by GENEBANK. At the base of pD-T23, a clone plasmid vector including TaGSkl, a pair of prime was designed to tag probe by PCR. Through Southern Blotting, TaGSkl was located in the chromosome of 1A1B1D (Homoeologous group 1) .Then according to the library of EST, TaGSkl was located in the short arm of chromosome of 1A, 1B and 1D.Glycogen synthase kinase acts as many important roles in many signal pathway. In order to study the role of glycogen synthase kinase in wheat in stress,this paper has studied the expression of glycogen synthase kinase in Triticum asetium(TaGSk1) in the stress of salt (treating time:0h, 1h, 3h and 12h), abscisic acid (treating time: 0h, 0.5h and 1h) and cold (4 °C centigrade)(treating time:0h,0.5h and 1h).In the way of half quantitative reverse transcription polymerase chain action (RT -PCR) .which actin is the inter-reference, the expression of TaGSkl increase under the stress. We also find that the expression of TaGSkl is higher in the salt-tolerance wheat RH8706-49 than in the salt-sensitivity wheat H8706-34.Along with the development of molecular biology, it is a new way to cultivate resistant salt wheat through gene engineering. This paper has settled the base of cultivating excellent quality wheat through gene engineering.
