Abstract: The wool follicles are the characterstic structure of ovine and all over the ovine skin.The mature of follicles is connected with more than twenty different cells origining from epidermis and hypoderma. The complex interaction among the cells and the effect of cell matrix will change the function of wool follicle cells, and the signal transduction of molecular mediating on the surface of cells.At the level of molecule ,It is beginning from synthesizing peculiar mRNA and protein for the development of ovine wool follicle. The radical reason is starting up of gene in the nucleus. It is a key point to understand the differentiation and development of ovine wool follicle from illustrating the gene expressive adjusting mechanism on level of moIecule.The selective expression of genes determines the development of ovine wool follicle.The molecule mechanism of development of ovine wool follicle can be discussed by looking for the correlative genes.Finally,We can analysis the biological process of controlling ovine wool quality.Firstly,The study sampled the Xinjiang Finewool and the Altay ovine as experimential animals,We studied ovine wool follicle cultural cells with 0.25% and 0.125% trypsin in vitro, the results showed that we had obtained two follicle cells of different shape. By contrasting and analysing the methods of isolating and culture, the results showed that heated digestion was better than cool digestion. While The Xinjiang Finewool wool follicle cultural cells were similar to the Altay ovine wool follicle cultural cells in shape.Secondly, The study sampled the China Merino Finewool as experimential animal,mRNA differential display reverse transcription PCR method (DDRT-PCR) was used to analyse and screen the development gene of the ovine wool follicles, During the experiment, the ovines were respectively injected 10ng/ml insulin-like Growth Factor(IGF-I), 20ng/ml epidermal Growth Factor(EGF), uninjecting growth factor after pulling out ovine wool and Control. There were three experiments and one contral.Three anchor primers and the eight arbitrary primers were selected. The follows could be done, RT-PCR,analyzing with electrophoresis,identificating and isolating the differential display bands, reamplificating ,cloning with pMD18-T vector, Northern blot analyzing, sequencing and searching by Blast software.The result showed SQ70(2) was compatible to EST sequence in cDNA library of bovine skin,Although we did not found the relative EST expression of IGF-I , EGF and their receptor.The identities were 95% and expect was 2e-58.It was showed that SQ70(2) was related to the growth and development of ovine skin (GenBank accession number:CX863390) as a known gene.While,SQ75(7) was a new gene in the ovine skin or a new high expressed EST in the ovine wool follicles (GenBank accession number:CX863391) for less than 90% Id.The whole sequence and the particular biological function of them will be studied subsequently.
