The Character Of MxMYB1 Gene Of The Genus Malus Xiaojinensis

There are many iron uptake relative genes cloned so far. But the majority of them encode transporter protein, few transcription factors are cloned. TaMER-BP fragment was used as probe for Malus xiaojinensis cDNA library screening and full length MxMYBl gene was obtained. MxMYBl expression was induced by iron. MxMYBl expression was undetectable by RNA gel blot analysis under iron sufficient conditions, it was induced greatly 3 days after being transferred to iron -deficient medium.Atlg70000 is a homologous gene of MxMYBl in Arabidopsis. Their amino acid homology is as high as 37%. Three T-SALK lines (salk₁43055, salk₀45905, salk₀25577) of Atlg70000 gene were analyzed. When T-SALK lines were grown on iron deficient and 300uM Ferrozine medium, they showed severe chlorotic, which could be rescued by the addition of supplemental iron. It indicated that these T-SALK lines are Fe-defect mutants. It indicated that MxMYBl and its homologous Atlg70000 gene might play an important role in iron uptake and mobilization.
