Peony is a traditional flower with great variety and wild resource. But the present preservation method is single and the lost of resource seriously. In this paper, cryopreservation procedure and physiological and physiochemical changes of peony pollen after and during LN2 storage were studied. Cryopreservation procedure was established, and the mechanism of cryopreservation was investigated. The results show:1. Cryopreservation can maintain peony pollen viability for a long time. Germination rate can still be kept well in LN2 storage after one year, and some cultivars exceed the original level.2. The results indicate: 10% sucrose+0.1% broic acid is a suitable medium for germination of peony pollen; pro-freezing at 3℃ and -30 ℃ are reduce the viability of pollen after LN2 storage; thawing in flowing water can enhance the pollen germination after LN2 storage. So the cryopreservation procedure of peony pollen is: collecting mature pollen—storage in LN2 without pro-freezing—thawing in flowing water—mensurating germination in 10% sucrose+0.1% broic acid medium about 25 ℃.3. Temperature of preservation affect pollen viability notably. 3℃ can keep pollen viability in 7 days; -30℃ can keep pollen viability well after one month; but cryopreservation is the only one effective method can keep pollen viability for a long time.4. The changes of physiological and physiochemical of peony pollen after and during LN2 storage showed: the membrane relative permeability increased constantly;the MDA content declined in the earlier days then raised with prolonging storage time; the speed of O2- production decreased notably;SOD activity raised gradually; CAT activity was not changed; the As A content first raised then declined after 30 days, afterward,it still raised gradually;the content of soluble protein and soluble sugar raised in the earlier stage then declined continually,the protein band don't changed after cryopreservation. In the cryopreservation course, physiological and physiochemical changes linked to cultivar difference,perhaps they had some effecton holding pollen viability.
