Study On The Genetic Diversity Of Germplasm Resources Cultivated In Ganoderma In China

151 strains of Ganoderma and 3 other strains were collected from different provinces of China and other countries, and their genetic diverdity were analyzed by the morphism of fruiting bodies , antagonistic reaction , RAPD (Random Amplified Polymorphic DNA) and ERIC-PCR ( Enterobacterior Repetitive Intergenic Consensus PCR) techniques respectively in the present study. The results were as follow:1. 133 fruit bodies were gained by cultivation, and by morphological characters, they were divided into ten species: Oligoporus guttulatus(Pk.)G'\\bn.Qt.Kyv is G0028; Ganoderma lucidum ( Chinese type), totaled 109 strains; G. sinense (type of thin hem and velvet spore) , did 5 strains; G. tsugae( Chinese type), did 3strains; G. tenue, did only one strain; G. resinaceium, did 5strains; G. subamboinense, did 2 strains; G.sp.(No.1), did only one; G. oerstedii, did only one; G. applantum, did 2 strains; G.sp. (No.2), did only one .2. The 151 Ganoderma strains were divided into 28 groups according to the presence or absence of the antagonistic line or curve ;3. The dendrogram derived from RAPD data indicated that (1) the above strains could be divided into four distinct groups and the result was in complete accordance with that of traditional classification at the low similarity level of 0.610; (2) some strains from Canada and America were different from those collected from China on the genetic relationship at the level of 0.670 and it indicated that the genetic diversity between them could be caused by geographical distribution; (3)those strains collected from China were divided into 25 groups at the high similarity level of 0.800 and the result was preliminary in accordance with that by antagonistic reaction.4. The dendrogram derived from ERIC-PCR data indicated that (1) the above strains couldbe divided into four distinct groups and the result was in complete accordance with that of traditional classification at the low similarity level of 0.670; and the relationship between GO 104 from China andG0128 from India was close and they were very different from the strains from America and Canada and it indicated that the genetic diversity between them could be caused by geographical distribution; (3)those strains collected from China were divided into 25 groups at the high similarity level of 0.800 and the result was preliminary in accordance with that by antagonistic reaction.According to these experiments, some reasons to cause the systematical chaos in thefield of Ganoderma and the feasibility and stability of different methods werediscussed in the present study.The genetic relationship between the prime cultural species of Ganoderma in Chinaand that between them and some cultures from other countries were first represented inthe present study, and it greatly helped to check the quality of Ganoderma lucidumcomplex in the course of production and study the their effects.
