Pathogentic Studies On Rotavirus Infection In Infant Golden Hamsters

Rotavirus is the leading cause of life-threatening diarrhea disease in infants and young animals worldwide, yet its pathogenesis remains uncertain. In this experiment the infant golden hamsters of natural rotavirus infection are used to study the natural history, clinical features, the structural and functional consequences of infection in multi-system, the tissues were sampled, fixed and stained for pathological observation, the peripheral blood were collected for some biochemical items, the frozen sections were cut for AKP, and the contact glasses of thymus and lymphonodi were pushed for examining T lymphocyte. The etiologic agent was identified to be group A rotavirus by means of EM and ELISA. The pathological changes showed the severe mucoid degeneration of intestinal epithelial cells, the presence of inclusion bodies in cytoplasma of cardiac muscle and intestinal epithelial, the degeneration, necrosis and edema of parenchymal organs, above all only the atrophy and fatty degeneration presence in cardiac muscle cells, and the hyaline degeneration exists in kidney. The hyperemia and hemorrhage are the main changes of lung. By electron microscope, viruses in clumps or sporadic were observed in intestinal epithelial cells.During infection, the content of red cell, white cell and hemoglobin in peripheral blood was descended sharply than normal, meanwhile the percentage of neutrophile granulocyte in leukocytes was raised, and the lymphocytes was reduced. The T cell rate in lymphonodi was extremely higher, and the small T cells in thymus was obviously lower. The AKP activity of the mucous membrane in intestine was higher too. The morphological changes of erythrocytes support anemia strongly. The conclusions:The Infant Golden hamsters are sensitive to RV;The cells of cardiac muscle and intestinal epithelial are objective cells to RV;The main pathological change was the mucoid degeneration of intestinal epithelial cells, and multi-system organs were involved.The infection of RV can cause the body's malnutrition, anemic, edema and low-immunity.The AKP activity of the mucous membrane in intestine was higher than one of the normal animal.
