The active elements from Salvia miltiorrhiza Bunge were isolated and structurally identified in the study. Ruyuankang injection for mastitis treatment had been made by the active antibacterial elements from Salvia miltiorrhiza Bunge and other Chinese herbal medicine. Its stability and pharmacological action had been evaluated. And then clinical effect of the injection had been observed through preliminary clinical application. The fat soluble chemical elements of Salvia miltiorrhiza Bunge were extracted by dipping in methanol, and isolated, purified by TLC, LSC and recrystallize. Four compounds were isolated from the roots of the herb. On the basis of UV, IR, 1HNMR spectra and m.p., and also by comparison with their spectral data reported in literatures, the structures were preliminary identified as follows: tansheninoneIIA, dansheninoneI, cryptotanshinone, dihydrotanshinoneI.Confirmed by a series of facts,the injection is steady. The antibacterial test showed that the injection could kill and restrain the pathogenic bacteria of mastitis, such as Streptococcus agalactiae, Streptococcus dysgalactiae, Staphylococcus aureus, Escheri-chia coli, Streptococcus lactis, Klebsiella. Eye-stimulating test in rabbit showed that there was no local stimulability for the injection. The acute toxicity reaction showed that the LD50 of Ruyuankang by intragastric transfusion (i.g.) was 36.80g/kgB.W,the 95% reliability was 32.12~41.47g/kgB.W.;the LD50 of Ruyuankang by intraperitoneal injection (i.p.) was 13.85g/kgB.W.,the 95% reliability was 12.76~14.94g/kgB.W. The results indicated that Ruyuankang injection had no acute toxicity for experimental animals. Assessing anti-inflammation function by dimethylbenzene-inuced ear oedema in mice,the result indicated that Ruyuankang might eliminate inflammation(p<0.05). The effect of anti-inflammation of Ruyuankang was same as hydrogen cortisone. One hundred twenty-two mastitis udders of 92 cows were treated by Ruyuankang injection, 28 and 34 udders among those were treated separately with penicillin and streptomycin, ciprofloxacin as control. The total efficiency was 94.7%, 93.7%, 93.2%,the total heal rate was 81.7%,70.8%,82.2%. The clinical results showed that Ruyuankang injection is safty and high heal rate for mastitis in dairy cows. Especially it could make the temperature normal and reduce udder lessening. This experiment firstly reported that combination recipe consisted of active elements from Salvia miltiorrhiza Bunge and other Chinese herbs which treated mastits in cows.
