Poplar is one of the important sylvicultural and commercial tree ,it is very popular throughout the world,especially in china. Populus euramericana 107 is a kind of hybrid strain from populus nigra and popolus deltoids. It is the main strain which has been popularized for its high values such as growing fast, high resistance, long fibre etc., which plays a very important role in reclamative afforestation and shelter belt industry. Chitin, a biopolymer of N-acetyl-D-glucosamine in a β-1,4 linkage, can be degraded to GlcNAC by chitinase. Many experiments show that chitinase shows strong inhibition to fungal growth both in vivo and in vitro. Cowpea trypsin inhibitor (CpTI ),which has a broad-spectrum anti-insect effect,can restrain the activity of alimentary canal proteinase in insect's body and retard the growth and development of insects, even cause the insects dead. In this study, we select the Populus euramericana 107 as the research material. Several factors such as hormone concentration were investigated to optimize the leaf regeneration system in vitro. A high frequency regeneration system in culture has been established. Based on this condition, two expression vectors containing CpTI gene and chitinase gene were constructed respectively,and we obtained some transgenic plants through the developed Agrobacterium tumefaciens mediated transformation method.
