Rice is very important in the plant structure of China. In recent years, the plant area of hybrid seeds enhanced greatly. It was very important to strengthen the purity assessment for rice planting. It aimed to set up an easy and effective technique in rice hybrid seeds purity assessment that could give some technique support to controlthe rice hybrid seeds purity. Based on previously research, it improved the method of AU-PAGE and SRAP and educed the protein electrophoresis technique that could be used in rice hybrid seeds purity assessment. The detail results were as follows: (1) It improved three aspects including the gel chroma, extract solution chroma and electrophoresis procedure, which could affect the electrophoresis result. Then, it established an easy and effective AU-PAGE technique system that could be used in variety identification and purity assessment for rice hybrid varieties and their parents. (2) Improved AU-PAGE was used to study the polymorphism of seed storage proteins in rice hybrid seeds. Albumins and glutelins showed more polymorphism than the other two compositions. Globulins did not gain the bands. The seed total storage proteins showed more polymorphism than albumins and glutelins, and the quality of its bands was very good. From the UPGMA dendrogrum of 17 different hybrid varieties, it found that the identification result was very good based on the seed total storage proteins and albumins. (3) It studied the diversity of seed total proteins of three rice hybrid varieties such as Liangyoupeijiu,â…¡you084 and Yueyou93B with improved AU-PAGE technique, and found that the rice seed total storage proteins compositions of parents all emerged in their hybrid seeds. With that technique, it tested the purity assessment of Liang-youpeijiu and it had a contrast with the result gained in field.
