Abstract: Chinese herbal drugs are rarity of traditional medicine,which effective constituents are polysaccharide,alkaloid, oil,et.al. Polysaccharide exist cells of animal,plant and microbe widely, it is one kind of basical material that make life,and participates in all kinds of cells'activities.From 70', scientists discovered that polysaccharides have powerful immune activity,from then on, polysaccharides become the center of Chinese herbal drugs pharmacological research especially immunity pharmacological research.Now,studies confirm that polysaccharides of Chinese herbal drugs have immunity regulate function,antiviral activity,anti-inflammatory, antioxidant effect, antitumor activity, anti-radiation effect, educing blood sugar effect,et.al.of which immunity regulate function is the focus of polysaccharides research.This study explore immunity regulate function of total Polysaccharide of Chinese herb recipe that maked by ourselves,and filter associated differential gene initially. Forty mice were randomly divided into four groups:normal saline group, Total Polysaccharide group, Astragalus Polysaccharides group and decoction group.After giving drugs continuously for 7 days,the mice were killed to detect the Lymphocyte transformation rate, the phagocytic functions of peritoneal macrophages(PM) and EA-rostte rate.At the same time,keep the splees to select differencial gene fragments by DDRT-RCR.Concludes are:1. Total Polysaccharide of Chinese herb recipe can advance specific immunology and nonspecific immunology obviously.Its effect is better than Astragalus Polysaccharides and decoction.So we can infer that Polysaccharide is the main content of immunity regulate function,and each polysaccharide from Chinese herb recipe operate together.2. In this experiment, total Polysaccharide of Chinese herb recipe can advance phagocytic functions of peritoneal macrophages(PM) and activity of Fc receptor, we can infer that Polysaccharide have antitumor activity that we will research in future.3.We get three differential fragments of mice by DDRT-PCR,and we found that LY3 homology is 99% compare to Mus musculus BAC clone RP23-20E4 from chromosome 3 by sequence analysis,L2 homology is 92% compare to Mus musculus BAC clone RP23-339H16 from 15,L3 homology is 98% compare to Mus musculus chromosome 7, clone RP24-371C6,We infer that these three differential fragments are contact with immunity regulate function of Polysaccharide.4.DDRT-PCR can select differencial expression gene from mice of total Polysaccharide group, Astragalus Polysaccharides group,which establishes the base of Polysaccharide research for the future.
