Dictamnus angustifolius Gdon belongs to Rutaceae. Cortex Dictami radicis is a traditional Xinjiang nationality material for medicine. It has the same medicinal efficacy as the root bark of Dictamnus dasycarpus Turcz. In recent years, it was researched and discovered that dictamnine and fraxinellone have the anticancer activity, and coarse polysaccharide can guarantee the liver and raise leukocytes. Because of artificial excavation and improper utilization, the wild resource of Dictamnus angustifolius Gdon was destroyed seriously. For plant rapid reproduce, the technique of plant somatic embryogenesis is one of the most important measure, which can obtain massive vegetative propagule in short time. It is significant to protect ecological system and exploit potentiality of Dictamnus angustifolius Gdon. Based on the above reason and the experience that has, we have done some initial work about somatic embryogenesis and protein expression of Dictamnus angustifolius Gdon in this thesis. The main work and results as follows:1. In the induction process of Dictamnus angustifolius Gdon somatic embryo, the 5th medium (KT 3.0 mg·L-1+NAA 0.1 mg·L-1 + MB+ CH 1000 mg·L-1)can induce embryogenic cells, and the 34th medium (6-BA 2.0 mg·L-1+ NAA 0.1 mg·L-1 + MS) can induce the somatic embryogenesis. On the other hand, we observed it by microscope at the same time. We found embryogenic cell is smaller than non-embryogenic cell, and the cell nucleus is bigger, and the cytoplasmic matrix is denser. It has the same growth process as most dicotyledons, and appears physiological isolation that could maintain independence.2. In the somatic embroygenesis process of Dictamnus angustifolius Gdon, the variation of POD activity is similar to SOD's, which correlates with globular embryoid and mature embryoid. The variation of CAT activity is related to mature embryoid. The DNA and protein synthetic rates have relations with the somatic embryo increment and tissue differentiation. Protein expression has existence certain difference. These special proteins could be molecular mark for appraising the latent somatic culture, so that those track the somatic embroygenesis of Dictamnus angustifolius Gdon. In the relevance of somatic embryogenesis of Dictamnus angustifolius Gdon, POD isozyme is more evident than SOD isozyme.The study, on somatic embryogenesis and protein expression of Dictamnus angustifolius Gdon, is significant scientific value and positive meaning to explore the somatic embroygenesis principle and exploit the distinguishing plant resources further, and protect ecological system.
