The present study was to improve the extender for frozen storage of buffalo semen and check some factors which influence the sperm performances by comparing some basic solutions, cryoprotectants and other additives.The results showed that the sperm motility was significantly lower inEYC than in the TRIS and TEST (P<0.05), but there was no significant difference between TRIS and TEST (P>0.05). There was significant difference (P<0.05) between TRIS and EYC at acrosomal and membrane integrity. The numbers of oocyte cleavages were significantly lower in EYC than in the other two solutions (P<0.05), but there was no significant difference between TRIS and TEST (P>0.05). The blastocyst rates was also significantly lower in EYC than in the other two solutions (P<0.01); The sperm motility was higher when quail egg yolk (QEY) was added as cold shock protectant than chicken egg yolk (CEY) (P<0.05) , but there was no significant difference in the acrosomal integrity between these two protectans. In the experiment of IVF, the cleavage rate and blastocyst hatched showed was higher in QEY group than in the CEY group (P<0.05). The sperm motility was higher in the group of 3ml/100ml the vitamine complex than in Oml/100ml groups (P<0.05) and the acrosomal integrity were no significant among groups (p>0.05). The sperm motilities were significant higher in the lmg/L and 2mg/L vitamine B12 groups than in the control (P<0.05) and also the acrosomal integrity was higher in the lmg/L vitamine B12 groups than in the control (P<0.05). The lmg/L the ascorbic acid could enhance the sperm motility and the acrosomal integrity when compared to the control. As the glycol concentrations as the cryoprotectant, 2% was better than 4% (P<0.01)and 6% (P<0.05), and 4% was better than 6% (P<0.05) in the sperm motility. A reversing phenomenon happened to the propanediol as a cryoprotectant. The sperm motility was significantly lower in 2% group than in the 4% and 6% groups (P<0.05). And the sperm motility was no significant difference between the groups of 4% and 6% (P>0.05). As the sperm motility, cleavage rate and the blastocyst rate, 4% glycol played a similar function as the 7% propanediol (P>0.05). 300IU/ml SOD could improve the sperm motility, livability and the acrosomal and membrane integrity rates (P<0.05). l.OmM GSH could also improve the sperm motility and prolong the sperm live span of stored semen (P<0.05). 300IU/ml SOD with l.OmM GSH was superior to only SOD or GSH in the sperm motility and live span (P<0.05). The results showed, in the test of antibiotics, that Gentamcin with Tylosin, Lincomycin, Spectinomycin was superior to Penicillin with Streptomycin (P<0.05) and Penicillin with Neomycin (P<0.01); Penicillin with Streptomycin was better than Penicillin with Neomycin (P<0.05).
