A high purity of hybrid rice seed is the guarantee of high yield, and a rapid, easy, economic and accurate method of seed purity test is the guarantee of supply of qualified seed. In this study, Shanyou63F1 , Teyou559 F1 and their male sterile lines and restorer lines were used to investigate the difference of incomplete leaf length between hybrid and their respective parents; and 6 hybrids, Teyou559, Ⅱyou559. Ⅱyou084, xieyou084, Fengliangyou 1, Liangyoupeijiu and their parents were use to study the polymorphism between hybrids and their respective parents amplified by SSR primers. The main results were as follows:1. Shanyou63F1, Teyou559 F1 and their male sterile lines, restorer lines were nursed with water. 5 days later, the incomplete leaf length(ILL) of all the 6 varities were measured. The mean of ILL was 14mm, and the maximum was 16mm in Zhenshan97A;The mean of ILL was 19mm, and the minimum was 17mm in Shanyou63F1.There was no overlap between the distribution of ILL in the 2 materials. Therefore ILL could be used as a indicator for seed purity testing in Shanyou63 whose parents were obviously different in plant height.However, under the same condition,there was overlap in the distribution of the ILL between Teyou559F1 and LongtepuA, and even they were treated with 1 mmol·L-1 GA3. so ILL couldn't be used in the seed purity testing of Teyou559 combination..2. SSR allele polymorphism of 6 hybrids and their parents was studied by analysing 40 SSR loci distributing on 12 chromosomes in rice. The results showed that SSR locus RM224 could discriminate F1 and their parents in all the 6 hybrids. Ⅱyou084 F1 and Fengliangyou 1 F1 could be identified from their respective parents by 3 pairs of SSR primer; Liangyoupeijiu F1 could be identified from its parents by 4 pairs of SSR primer. RM224, RM296 and RM282 can distinguish Ⅱ you084 and its parents. RM224, RM296 and RM489 can discriminate Fengliangyou 1 and its parents, RM224, RM296, RM282 and RM7 can discriminate Liangyoupeijiu from its parents. A sample of Liangyoupeijiu Fl mixed with parents on purpose was examined for its purity by using the primers of RM224,the testing result was consistent with the mixed ratio beforehand.
