Presently, microsatellites (MS) are widely introduced in parentage testing. MS are equably distributing in genome, high PIC, easy to detect and repeat, and suitable to automatization analysis. So it is very important in individual identify (including parentage testing), linkage analysis, population genetics studying. Based on the enhanced linkage map of the sheep genome, we selected out 20 microsatellites BL0004, BM1577, BM2830, BM3501, BM6465, BM6506, BMC1009 BMS0887, BMS1636, BMS1678, BMS2321,BMS2626,ILSTS004, ILSTS0049, ILSTS008, ILSTS22, OarCP0034, OarCP43, OarFCB0226, OarFCB20 on Chr1, Chr2, Chr3, Chr9 of sheep for studying in LiangShan semi-wool sheep. And we experimented with fluorescence primers, multiple polymerase chain reaction and ABI310 capillary electrophoresis for paternity testing, in order to expose real relative of the subject and construct an exact family tree of the local variety.Parentage exclusion probabilities were calculated from allele frequencies in 353 individuals. The combined exclusion probability is 0.998666 when parents are both unknown. If father is known, combined exclusion probability is 0.999994. Finally, we found the biological parent of 215 offspring form 8 candidate fathers and 147 candidate mothers, and constructed 8 pedigrees of sire family and a mixed pedigree. These are useful for linkage analysis of sheep chromosomes and QTL locating, also for studying of population structure, history, and diversity.
