Powdery mildew is one of the most serious diseases affecting wheat yield.InChina,one of important question in wheat breeding of resisting powdery mildewis single resistant source.For the reasons of needing specific method ofinoculability identification and affected by ambient conditions,the results ofresistance identification of powdery mildew were trustless frequently.It results inthat lots of effective resistance genes have not been used as fully as possible.Withextensive application of molecular marker assistant breeding in wheat cultivargenetic improvement,it has important significance to build effective molecularmarkers of resistance genes for using molecular marker assistant selection toimprove efficiency of wheat resistance genetic improvement.In this study,twomolecular markers were identified to be linked to in coupling phase to Pm12 andPm3b by using RAPD technique.The results are showed just as follows. 1,Cross was carried out between Pm12 as resistant parent and Chancellor assusceptible parent,and F1 plants from the cross were selfed to obtain F2 mappingpopulation.Bulked segregant analysis (BSA) was used to search for RAPDmarkers linked to the Pm12 gene,which confers resistance to the powdery mildewin wheat. 230 decamer primers were screened and identified one RAPD marker(S1071900) linked to in coupling phase to Pm12.Using the F2 mapping populationto make RAPD analysis,Pm12 was shown to be closely linked to the markerS1071900 at a genetic distance of 11.98±4.00cM. 2,Cross was made between Pm3b as resistant parent and Chancellor as susceptibleparent,and F1 plants from the cross were selfed to obtain F2 mapping population.BSA wasused to search for RAPD markers linked to the Pm3b gene,which confers resistance to thepowdery mildew in wheat. 230 decamer primers were screened and identified one RAPDmarker (S2181100) linked to in coupling phase to Pm3b.Using the F2 mapping population tomake RAPD analysis,Pm3b was shown to be closely linked to the marker S2181100 at agenetic distance of 11.22±3.56cM.
