The potato tuber dormancy influences the production and consumption of potato. It is the most important to resolve in potato production how to control potato tuber dormancy maintenance and release. The experiment analyzed changes of potato microtuber and minituber protein in different periods that were stored at different temperatures by SDS-PAGE. We also studied the physiological and morphological changes during tuber dormancy maintenance and release. We combined the protein stain with DNA stain to obtain excellent stain results, which can provide a new method for protein stain. New protein bands appeared when sprout occurred. These polypeptides maybe associate with dormancy release of potato tuber. It laid the groundwork for deep research on mechanism of dormancy and foundation for industrialized controlling dormancy system. The results are as follows:1 B9 made stems and leaves of virus-free plantlets stronger, different concentration having different results. The MS medium supplemented with B9 3 mg/L was suitable for strong sprout by analyzing effect of B9 on virus-free plantlet of potato in vitro.2 System of SDS-PAGE was optimized, conditions being running gel 12%, gel thickness 0.75 mm, and loading amount 10 ul.3 High resolving power method for potato protein-improved silver stain method was established.4 SDS-PAGE eletrophoretic patterns of protein of both potato microtuber and minituber were consistent in different periods that were stored at different temperatures. Two polypeptide bands of alcohol soluble protein and one band of soluble protein appeared when sprout came. It was time to sprout for the potato eye. These polypeptides may be associated with dormancy release of potato tuber because it was time to sprout for the potato microtuber and minituber.5 Alcohol soluble protein and soluble protein content of'Zhongshu 3'microtuber changed a little bit during storage. At the 6th week, soluble protein increased greatly; at the 16th week soluble protein concentration reached maximum, 2.152 mg/g; at the 18th week alcohol soluble protein concentration reached maximum, 1.448 mg/g. The rapid increase in the content of tuber protein at the 6th week would be a signal for dormancy release. 6 Temperature affected duration of potato microtuber dormancy. At 4℃, sprouts in microtubers was suppressed and grew slowly. Low temperature kept duration of dormancy longer.
