Typical goose Newcastle disease outbreaks in three goose farms in Shandong Province in 2003. The epidemiological results showed that the goose ND could occur in different ages geese and species, but the incidence rates of young and purebred goose were higher than that of adults and breeding. The incidence rates in the three goose farms were 100 %,90% and 80%respectively, and the mortality rates is 75% , 70% 55%respectively.The samples of infected geese brain and spleen were collected, treated and inoculated 11-day-old SPF chick embryos and 16-day-old goose embryos. A ND virus (named strain WF216) was isolated from the dead chick and goose embryos. The isolate could agglutinate chick erythrocyte, and could be inhibited by the standard antiserum of the NDV, but it couldn't be inhibited by the standard antiserum of the AIV H5N1 and EDSV -76. It can be concluded that the isolate was goose ND virus.The virulence of the goose Newcastle disease virus strain WF216 was identified according to the standards recommended by OIE. The results showed that the mean death time(MDT) in chick embryos is 49.9h, the intracerebral pathogenicity indices(ICPI) in one-day-old chick is 1.65, and the intravenous pathogenicity indices(IVPI) in 6-week-old chick is 2.23. It can be concluded that the virus of strain WF216 is high virulent strain.Twenty unimmunized chicken and geese of 25-day-old were inoculated with goose Newcastle disease virus strain WF216 and the contrast group with physiological salt. As the result, all the tested chicken and geese showed typical clinical symptoms and pathological changes of ND and goose ND. It suggested that strain WF216 is identical to the pathogen of natural goose ND.Ten-day-old unimmunized chicken and geese were inoculated with the inactivated oil-adjuvanted vaccine made from the isolated goose ND virus strain WF216 and the contrast group with physiological salt. 21 days after vaccination, each chick and goose in test and contrast group was injected individually 0.1ml 10-1 diluted fluid of goose ND virus strain WF216. As the result, chicken and geese in contrast group showed typical clinical symptoms and pathological changes of ND and goose ND. On the contrary, all the tested chicken and geese kept alive and healthy.Ten-day-old unimmunized chicken and geese were inoculated with the NDV inactivated oil-adjuvanted vaccine and the contrast group with physiological salt. 21 days after vaccination, each chick and goose in test and control group was injected individually 0.1ml 10-1 diluted fluid of goose ND virus strain WF216. As the result, chicken and geese in contrast group showed typical clinical symptoms and pathological changes of ND and goose ND. On the contrary, all the tested chicken and geese kept alive and healthy.The tests indicated that both the inactivated oil-adjuvanted vaccines made from the isolated goose ND virus WF216 strain and that from NDV can exempt chicken and geesefrom the attack of wild goose ND virus.Healthy hens were vaccinated with the inactivated oil-adjuvanted vaccine made from the isolated goose ND virus strain WF216 to produce high immunized IgY of goose Newcastle disease virus. Treatment test and preventive test have been processed.The treatment test showed that the inactivated oil-adjuvanted vaccine can treat the geese that were infected artificially by goose ND virus( 17/20) effectively, and so does the natural infected geese. The cure rate was over 80%.The preventive test showed that each tested goose injected goose ND virus high-immunized IgY by the dose of 1 .0ml. After 24 hours, each goose in test and contrast group was injected 0.1ml 10-1 diluted fluid of goose ND virus strain WF216. The result showed that all the tested geese kept alive while all the contrast ones died. |
