The Study On Rhizosphere Bacteria Of Transgenic Xa21 Rice

The research was comparing with the variety of three different bacterium communities and the inspection of significance difference notably. The bacterium communities regarding Shannon's index (Shannon index ) as community's diversified index were from the rhizosphere of three rice varieties (transgeneic rice of Xa21, Wan A, Xianyou 63).Districts of arranging at random were designed respectively in Hubei Academy of Agricultural Sciences experimental plot and Chinese Academy of Agricultural Sciences Institute of Plant Protection experimental greenhouse .The experimental result indicated there wasn't the significance difference between the six treatments in field and greenhouse on rhizosphere bacterium community structure and population's figure .The bacterium community population's figure of six treatments in greenhouse and field were changed with change of season, which meaned there were similar changes in microorganism's population in the course of the crop's growth ,it indecated the bacterium's adaptability. TR (transgenic rice ) was far smaller than the influence of season , climate , area to the bacterium's community's structure of rhizosphere.The sequence of intergenic transcribe spacer region(ITS, ITS is between the area of 16S rRNA and 23S rRNA) of advantage bacterium in rhizosphere was cloned and compared with array.The results had obvious difference on the length of ITS between several important genus, the result are respectively A 745bp, B 377bp,C 329bp,D 574bp, E 377bp,F 647bp,G 319bp, H 352bp, I 495bp.According to the array of the report,the nine separated advantage bacteriums belong to five different bacterium genus,Which are Bacillus, Flexibacter flexilis , Xanthomonas, Escherichia coll ,Tropheryma ,Then we got the dynamic chang of different bacterium genus .Utilizing ITS can be fast qualificate the bacterium !As origin centre of the rice,it is very important to our country to protects the hereditary resource of rice , bio-diversity and the security of the transgenic rice, especially the ecological security , so it have very important meaning to study the ecological security of transgeneic rice ofXa21 for promoting the commoditization of the transgenic rice of our country.
