Studies On Immune Colloidal Gold Diagnostic Technique Of Newcastle Disease

Developing a rapid kit for the diagnostic of Chicken Newcastle disease Chicken Newcastle Disease is a highly infectious and deadly disease, it is caused by NDV (Newcastle disease virus) that violate the respiratory tract the digestive tract and the centrencephalic system. The viruses are in blood, brain, internal organs excrement mouth and nose, there is no complete solution for this disease at home and abroad, so it is a big menace for chicken cultivating industry. So it is necessary to establish a rapid, simple,self-performing GICA(gold immunochromatographic assay) for the diagnosis of Chicken Newcastle Disease. Colloidal gold technique firstly was a kind of the tracking technique , which was reported to apply to electron microscopy(IEM) detecting technique by persons like Faulk in 1971. By comparison with the traditional technique, since the colloidal gold embodies the property of red to be seen by nake eyes, so the detective test box developed basically on this technique has rapid, sensitive, special, sensitive features, colloidal golds were coupled with Chicken Newcastle Disease to prepare an GICA apparatus, marking the purified antibody of Chicken Newcastle Disease, wrapping them onto nitrocellose strips, and then wrapping themand the purified rabbit antibody for chicken onto nitrocellose, assembled a rapid kit for the diagnostic of Chicken Newcastle Disease. If the sample contains virus antigen of Chicken Newcastle Disease and the kit appears two red lines, it is positive; if the sample contains no virus antigen of Chicken Newcastle Disease and the kit appears one red line, it is negative. When using the rapid diagnosis of kits for Chicken Newcastle Disease, which are developed in this experiment, to detect the accumulated 20 samples, their results correspond the result of HA, and the corresponding rate is 100%, When virus HA titer is above 1:32, the kits are all positive. The rapid diagnosis of kit for Chicken Newcastle Disease appears no intercross reaction with the clear liquid in Avian Ingluenza(AI) and Chicken Infectious Bronchitis(CIB). It was confirmed that the GICA(gold immunochromatographic assay) established in this experiment is a kind of rapid, senstive, and special antigen test method, it can be used to test Chicken Newcastle Disease, and to investigate epidemiology of Chicken Newcastle Disease, and has the value to be generalized.
