| It is successful to use optimum SDS method to extract Cynodon dactylon(L.) Pers. general DNA, and the extracted general DNA of Cynodon dactylon(L.) Pers. is suitable for molecular biology study. The optimum RAPD reaction system in Cynodon dactylon(L.) Pers was settled. One hundred decamer oligonucleotide random primers from Operon Technologies were used for polymorphic selection, and 10 primers were selected for RAPD analyses. The date from 10 primers was used to generate genetic similarity and to construct a dendrogram. The main results show as follows:1) One hundred decamer oligonucleotide random primers from Operon Technologies were used for polymorphic selection. 36(36%) produced polymorphic RAPD products, and10 primers selected for RAPD analysis produced 116 amplified fragments and total percentage of polymorphic loci is 93.10. 9.25% polymorphic loci were different between wide populations and culture populations. There were characterized polymorphic loci for ten populations. Those result reveal high variation among sixteen populations.2) The genetic similarity for sixteen populations ranged between 0.3457 and 0.8611. The sixteen populations can be clustered into two groups when the similarity coeffients of 0.58 are used as critical value. Four culture populations cluster into one group. Twelve wide populations cluster into another group. Results of cluster analysis indicate that relationships between the twelve Cynodon dactylon(L.) wild populations are related to their geographic distribution. Compared with the twelve wild populations, difference among the four culture populations is smaller. This result is related to the influence of human beings to these culture populations. |
