| On the base of obtaining transgene Shiva-I regenerated plants produced by Agrobacteria-mediated transformation, the study were made on the expression and inheritance mode of foreign gene in the progenies, the altitude and phenotype of the transgenic and nontransgenic Paulownia plants at seeding and grow-up stage.The objects were the three independently transgenic lines of maternal plants RO, their root-vegetative propagation R1 progeny, the selfed progeny F1 and crossed progeny F2. The Polymerase Chain Reaction (PCR), dot blot hybridization and Reverse Transcription PCR (RT-PCR) were used to examine and analyze the transgenic Paulownia. The result showed eight examined random samples from RO group and seven from RI group expressed the gene Shiva-I; the segregation ratio of positive to negative plants in FI group fit the Mendel's Law (3:1), so did the F2 (1:1). The analysis of Dot blot hybridization and RT-PCR confirmed the result. The above preliminary result showed foreign gene Shiva-I expressed in the RO and R1 and the expression and inheritance mode in the progenies of transgenic Paulownia accorded with the Mendel's Law.The study on the altitude and phenotype at grow-up stage showed at earlier stage transgenic maternal plants and nontransgenic plants had no difference at altitude, but at later stage the former grew slower than the latter did and they had some difference. As time went by the difference had tendency to increase. A few transgenic plants flowered and had seeds in the same year.The study on the altitude and phenotype of plants at seeding stage showed three FI progeny transgenic lines expressed some difference at growth rate. But in the same period they showed no difference at altitude. They were shorter than the control and showed obvious difference. On the way of individual growth, the altitude of transgenic Paulownia was inhomogeneous greatly. The survival rate of transgenic and nontransgenic Paulownia declined. Three transgcnic lines expressed sonic difference at the survival rate, and their survival rate were lower than control's. On the surveying of phenotype buds were found on five transgcnic plants, but they didn't blossom. Before transplanted into nursery the five plants died.The study provided some reference basis for the application of transgenic technique in genetic melioration and some theoretical basis for the application and popularization of transgcnic Paulownia. |
