| Adipocyte function uniquely to accumulate adipose. The metabolism and construction features of adipocytes has been well investigated for years so as to understand the regulating mechanism. With the help of SDS-PAGE, Western-blotting and monoclonal antibody, it has been found that there are differences in the components of adipocyte membrane protein among different animals. Cell membrane is important for such processes as information transmission, nutrient transport and hormone signal identification. Therefore it has been speculated that there are some components of membrane proteins different from those in other tissues, which are essential for the deposition of adipose in adipocytes. The deposition of adipose in animals and the growth of adipose tissues result from the accumulation of lipid droplets in adipocytes and their continuous differentiation. Excessive deposition of adipose affects not only the product traits of livestocks but also the health of human beings, which has become one of the main problems in livestock cultivation and public health. Thus it is of great significance to study the components of membrane proteins and identify the specific membrane proteins of adipocytes in order to make insight into the structures and functions of the membrane proteins of adipocytes and search for satisfactory methods to decrease the deposition of adipose.In this study the membrane proteins were prepared with density and gradient centrifugation of saccharu from various animals, different swine species and different adipocytes and different tissues and organs of the same swine species. Theconstitution, molecular weight and content percentage of membrane proteins were determined with SDS-PAGE and thin layer scan. The specific membrane proteins in swine subcutaneous adipocytes were identified with Western-blotting. The results showed that there were about 18-22 kinds of membrane proteins in the adipocytes of pigs, bovine, sheep, chickens, rabbits, mice and human beings, of which 6 kinds of membrane proteins of 110, 94, 65, 54, 44 and 35 kDa were present in adipocyte membranes of all animals. And the membrane proteins of 151, 80 , 144, 89, 76, 86, 55, 85, 61 , 99, 71, 23 , 116 and 82 kDa were in adipocyte membranes of different animals. There were great differences in the components and contents of adipocyte membrane proteins among different animals. 18-22 kinds of membrane proteins were found in adipocytes of .different swine species of Diankun, Dianyu, Dianlu, Changsa, Yuechangsha, DLY, Saba and Changbai pigs, of which 12 membrane proteins were of the same kinds. Their molecular weights were 110, 102 , 94, 72, 65, 55, 52, 50, 40, 36, 30 and21kDa respectively. The membrane proteins of 85, 138, 37 ,31 and 24 kDa were found hi adipocytes of different swine species. The components of adipocyte membrane proteins of different swirie were similar but the contents of each protein were different.19 kinds of membrane proteins were found in subcutaneous adipocytes of Changsa pigs. 13-24 kinds of membrane proteins were located in the cells of heart, liver, spleen, lung, kidney and skeletal muscles and in the adipocytes of renal, intestinal and abdominal adipose. 6 kinds of membrane proteins were in the membrane of erythrocytes. 8 specific membrane proteins were found in subcutaneous adipocytes identified with Western-blotting. The molecular weights were 110.7, 104.0, 40.0, 31.1, 29.4, 25:4, 23.5 and 22.0kDa respectively. There were great differences in the components and contents of membrane proteins between adipocytes and other cell membranes.The specific membrane protein of 40Kda in swine subcutaneous adipocyteswere purified and then the rabbits were immunized with it by multi-points injections. 8 weeks later the blood Was collected and antiserum with high titer was prepared. |
