| Five coding protein gene were studied in four China endemic pig breeds, Erhualian, Huaizhu, Zangzhu and Jiangquhai, by means of PCR-SSCP and PCR-RFLPs.The five gene loci were growth hormone (GH), Ryanodine receptor (RYR1), Leptin, hormone sensitive lipase (HSL) and Pituitary transcription factor (PIT-1). The results showed that there were polymorphisms at GH, RYR1, Leptin, HSL, PIT-1.two HhaI polymorphism were abundant in four breeds.No negative Halothane genotype was found in four China local pigs, a SSCP mutation was found in EHL and ZZ at Halothane gene. HinfI. polymorphism were found in EHL, JQH and ZZ,not in HZ,about HSL ,MM genotype was dominant in four pigs, A MspI polymorphism, different from American breeds was found in local breeds. Variance analysis showed the significant correlation (p<0.05) of the SSCP polymorphism of RYR1 with litter size. Other loci were not significant, The Nei's gene distance was computed by using the procedure GENDIST in PHYLIP package, and cluster tree data was computed by means of UMPGA and NJ using NEIGHBOR procedure, and the tree was drawn using TREEVIEW procedure. The result showed that HZ and JQH (UMPGA) or EHL (NJ) was the nearest within four breeds, and ZZ was farest, the results accorded to their geography distance. The article also conducted the problems existing in species conversion. |
