Study On Separation And Biological Activities Of Phenolic Compounds In Different Varieties Grape Skins

To develop Sephadex LH-20 chromatography method separated and purified phenolic compounds of grape skins in Shang-24(V.quinguangularis rehd), Tonghua-3 (V.amarensis Rupr)and Cabernet Sauvignon (V.vinifera L),and 8 fractions of 5-12 tube number was collected. To establish four bioactivities indexes (the capacity to scavenge superoxide anion radical (O2 -);scavenge hydroxyl radical (HO );chelate ferrons II (Fe2+);inhibit oxidized low-density lipoprotein(ox-LDL)) determined the bioactivities of every fractions.The difference among different mature stages, different varieties, and different bioactivities indexes were studied. At last, the results are as following :(1) The significant difference was found between four mature stages .The bioactivities in high mature stage is far higher than in low nature stage, and the highest bioactivity was found at forth mature stage in Shang-24,in Tonghua-3 at third stage, and in Cabernet Sauvignon at forth stage.(2) The significant difference was also found among different varieties. Compared with Cabernet Sauvignon, the bioactivities of phenolic fractions from Shang -24 were quite high. At forth mature stage ,the capacity to scavenging O2 ""of NO. 9 fractions from Shang-24 was 3 fold Cabernet Sauvignon's; scavenging HO of NO. 8 fractions from Shang -24 was about 5 fold Cabernet Sauvignon's; chelating Fe2+ of NO.7 fractions from Shang-24 was about 6 fold Cabernet Sauvignon's; and inhibiting ox-LDL of NO.8 fractions from Shang-24 was the same level as Cabernet Sauvignon's. The results showed bioactivities of Shang-24 grape skins phenolic compounds were the highest, Tonghua-3 were high, and Cabernet Sauvignon were the lowest.(3) The bioactivitive intensity of phenolic compounds in every variety grapes skins appears a peak, four fractions (NO.6,NO.7,NO.8,NO.9) were higher peak ranges.(4) The bioactivitive intensity from the same fractions had no identity .Certain fractions had higher bioactivity in one certain index ,and had lower bioactivity in another index . For example, at forth stage, NO.9 fraction from Shang-24 had the highest bioactivity in scavenging O2 capacity, the lowest bioactivity in scavenging HO , higher bioactivity in chelating Fe2+ and inhibiting ox-LDL . This further confirmed the structure-activity relationship of phenolic compounds.( 5 ) Under the condition of this text, the fractions with the highest bioactivity were: NO.9 fraction from Shang-24 grape skins at forth mature stage in scavenging O2 " capacity; NO.8 fraction from Shang-24 grape skins at forth mature stage in scavenging HO -capacity; NO.7 fraction from Shang-24 grape skins at forth mature stage in chelating Fe2+capacity; NO. 8 fraction from Tonghua-3 grape skins at third mature stage in inhibiting ox-LDL capacity.
