By means of techniques of cell fusion, the spleen cells of BALB/c immuned with sporoziotes soluble antigen of E.acervulina four times after, were fused with SP2/0 myeloma and screened through indirect ELISA to gain two hybrid cell strains which are 5B4 and 5G7 cell strains able to secret specific antibody steadily.Monoclonal antibodies from ascites were purified through combination of caprilic acid and ammonium sulfate and measured by indirect ELISA, the results before purified and after purified have shown McAb 5B4 is 1:131072 and 1:65536, McAb 5G7 1: 262144 and 1: 131072 respectively; the results of SDS-PAGE had shown way of purifision is suitable for two cell strains. Monoclonal antibody cell strains produced previously, were characterized by means of chromosomes segregation and analysis of hybridoma, immunofluorescence technic(IFT), immunohistochemical method and subclass of antibodies.The results had shown that two cell strains each of whose the average of chromosomes was close to the sum of chromosomes of two parent cells; two monoclonal antibody cell strains produced are specific for sporozoites of E.acervulina. Subclass of all two is IgGl.
