Wheat leaf rust is one of main important diseases which has caused heavy losses to the wheat production in the world. Use of resistant cultivars has been considered as an effective and important measure for preventing production from the damage caused by the disease. Understanding the physiological differentiation of the wheat leaf rust had very important guidance in the distribution of the resistant cultivars.AFLP is a new effective method of molecular marker. It was mostly used on the research of high mammal and plants. The fingerprinting analysis based on AFLP was carried out in 60 different wheat leaf rust strains coming from different areas. The virulence polymorphism and molecular polymorphism were compared in this paper, and the main results were as follows:1. A most effective AFLP technological system was established. Some improved factors were below:(1)restriction time was 3 hours; (2)the most available condition of ligation reaction was 22癈, 3 hours and 16癈, 10 hours; (3)the products of ligation were diluted to 5 times for preamplification; (4)the products of preamplification reaction were diluted to 5 times for selective amplification.2. With 12 fixed identifying gene, 17 assistant identifying gene and Thatcher, the virulence of 60 strains of wheat leaf rust were identified. Virulence similarity coefficient was between 0.58 and 0.97, and virulence difference has some relativity with region resource.3. Using AFLP method, 62 out of 64 pairs of primers showed effective in the analysis of molecular polymorphism. Similarity coefficient of molecular genetic was between 0.74 and 0.87. In addion, the molecular polymorphism was very relative with region resource.4. Through comparing the polymorphism of virulence and molecular genetic, it was found that the two were some relative but not relative completely.
