| Chinese sorghum ,Setcreasea pallida Rose and maize DNAs were introduced into common wheat varieties included jining13 hao,laizhou953, jihe916,yangmai158, 8017-2,jinan17 hao. Through the qualification of salt and drought resistance of wheat variation offsprings produced by exogenous DNA introduction, four variations off springs were screened with better salt and drought resistance. At the same time , differences between four variation off springs and their receptors in characters, protein,esterase and DNA were learned. The main results were as follow:1 Through the qualification of salt and drought resistance indoor and greenhouse, four were screened with better salt and drought resistance, such as 1174,1175 and 11488,11315.Their drought resistance grades indoor are first and salt resistance grades indoor are second. The drought and salt resistance grades in greenhouse are second. All of their resistances are more excellent than the receptor. Also the root length and number were thought to be able to act as the standard of the drought resistance . But their value should be learned further.2 The physiological index results indicated that the terminal contribution and increase every day of the solute sugar of four high-salt-resistance offsprings were more than the two receptors. Then it was deduced that salt resistance were superior to their receptors. Also the proline contribution results suggested that the two receptors' contribution and increase were more than the four offsprings. Hence the fours' salt resistance and superior than the receptors. But it was also thought that neither the solute sugar nor the proline could be looked as the sole salt resistance standard and they shoud be used with other index .3 RAPD result with the primer S317 indicated that four variations offsprings all had two exceptionally different belts from the receptor. Four variations offsprings and the donor had the exceptionally different belts which molecular weight was 650bp, but not the receptors. This probably wasDNA's part conformity in the maize gene group to the gene group of receptor. Also the exceptionally different belts appeared simultaneously in all of the primers' RAPD result, which proved that the sequences of exogenous DNA acted on certain "Hotpoint" of receptors' genome.4 The result of seed gliadin electrophoresis showed that there were differences in zymotye between the four variations off springs and the receptors, which was appearance of new belts and disappearance of the receptors'. It was verified that the injection of exogenous DNA influenced the components of gliadin. 5 The result of esterase electrophoresis showed that 1174 and 1175 didn't have the special belt existed in laizhou953.So it was concluded that the variety of esterase was the result of hereditary substance change and gene silence. There was no difference between 11315,11488 and their receptors . 6 The part of agronomy characters investigation showed the four wheat variation off springs were superior to the receptor. Their stalks were shorter and average stalk yields, which could be regarded as the yield-predicting standard, were superior to their receptors. Then it could be concluded that their yields every hm2 should be higher than their receptors and have good prospect in the production in drought and salt zone. |
