Plasmid GH includes the geneome of PPV NADL-2 strain . One pair of PCR primer for PPV VP2 gene wasdesigned according to the DNA sequence of PPV NADL-2 strain and designed by BamH I /Not I .With the primer for ,DNA fragment (about 2.2Kb) contain complete VP2 gene except 366bp before ATG of VP2 gene. The amplified fragment was purified and then cloned into plasmid PGEM-T-EASY, named the recombinant plasmid PPW.Restricted endonuclease analysis confirmed the accomplishment of the amplification and cloning of VP2 gene of PPV NADL-2 strain. Plasmid PPW was digested by BamH I /Not I ,purfied and subcloned into plasmid pusk ,which includes the gG gene of Ea strain, to produce recombinant plasmid PUSP. The resulting transfer vector PUSP will be useful for developing gG-deleted recombinant Pseudorabies virus to express PPV VP2 gene.
