In the man-made condition ,the Tobacco Budworm, Helicoverpa assulta Guenee, were induced diapause from eggs. Observing the molting, we divided the instar basing on it. Then extracted the RNA of different instar larves , pupas and moths. By using it as the model to synthesize the first strain cDNA. Aimed for the better result we designed the two pairs of primers for the PCR, studying the expression of the DH gene on the transcription. The result of the PCR showed that the DH gene expressed in the larves of the thirth instar,the fourth instar,the fifth instar,the sixth instar and the pupas. And it did not express in the first instar and the second instar.To know whether the DH gene could express in the pupa after the diapause terminate, the pupas and the moth of which the diapause terminate were studied by using the same method as above. The result or RT-PCR showed that the gene also could express in the terminated diapause pupa,yet it did not express in the moths.Dissecting the tobacco budworm,we separated the brain,thorax,and abdomen. The RT-PCR result showed that the brain was the special part in which the gene expressed.It was the first time to study the timal and the spacial expression of the DH gene in the insect of pupa diapause. The expression of DH gene is not only related to the development stage, but also related to part.
