Directed and indirected methods were used to study somatic embryogenesis of Cucumis melon L.cvHetao in this experiment, in directed method, we considered factors such as hormone, induction time, dark culture time, the physilogical condition of explant, carbohydrate source that affect somatic embryogenesis. As a result, hormone played a key role, its composition and concentration were very important in induction of embryogenetic.High concentration Glucorse apparently enhanced embryogenic rate. Regenerated plants were achieved by directed method. So we builded a plant regeneration system by directed somatic embryogenesis. Based on directed method, far more research was done by indirected somatic embryogenesis method, as a result, hormone, basal medium, activated charcoal, inositol and CH had apparent effect on indirected somatic embryogenesis. Smooth-transparent and white-tight embryos were achieved,by paraffin-slice observed, they were ellipsoid developed-stage embryos.
