B lymphocytes were got from BALB/c mouse which have been immuned with pure recombined antigen of Xinjiang Hemorrhagic Fever (XHF) virus beforehand, then those B lymphocytes were fused with myeloma cell-SP2/0 by PEG. After positive clone selection and monoclonal cell culture, 10 strains monoclonal antibodies (McAb) against recombined XHFV nucleoprotein antigen were obtained. Mouse monoclonal antibody kit was used to make pure McAb, mouse antibody isotyping kit was used to check the type and isotype of those McAb. After detection, 6 IgM type and 4 IgG1 type McAb were identified, and the all IgG type McAb can capture XHFV antigen with antigen capture ELISA but only 2 of the 6 IgM McAb did.Because of the serious outbreak of XHF in Bachu County in 2001, we completed the seroepidemiologic investigation of XHF from middle May to the end of June in the area. Serum samples from XHF patients, normal peasants, and sheep (goats) were collected, and special IgG antibody and IgM antibody against XHF were checked by indirect ELISA, XHFV antigen was checked by antigen capture ELISA, which was established by McAb we have got. In addition, XHFV nucleic acid was checked by RT-PCR. The results showed positive rate of antibody was very high and there were some recessive infections in peasants and sheep (goats).
