Separate And Identify Cold-induced Gene In The Chinese Straw Mushroom, Volvariella Volvacea

Volvariella volvacea (Bull.ex Fr.)Sing..commonly known as the Chinese Straw Mushroom is an important edible fungus of thetropics and subtropics.Chilling injury also has been found in this mushroom. Low temperature induced V.Volvacea cell protein degradation and result in myceliaum autolyzed. Try to understand the physiological change in cold autolaysis of V.Volvacea proteinase cold induced protein and cold induced genes have been investigated.RNA differential display was used to investigate the cold shock gene expression 19 random chose primers combined with 4 Oligo primers [Oligo-p(dT)i5A, Oligo-p(dT)|3C, 01igo-p(dT)nG. Oligo-p(dT)|4] were amplifyied 63 cold-shock DNA fragments in 32 18-25 bp random chose primers. DIG labeled cDNA probe was synthesize by PCR. The result of cDNA- DNA hybridization show that 12 DNA fragments were special cold-induced DNA fragments. Hybridize digested total DNA of V.Volvacea with 12 cold-induced DIG-DNA probe.The result indicated that cold-induced gene was low copy in genetic DNA of V.Volvacea .
