Elymus.rectisetus, the loose poly-annuais grass family, is the only apomictic reproduction within the tribe triticease. It belong to the type of 2n-pollen-producing. Exception of the apomixis it also has the good property of resisting pathy^ cokU drought salt and base, it also contain high protein. So it's the good species source of wheat. This research using thirteen selected random primer (OPE-09, OPF-O5, OPF-09,OPN-12, OPE-ll,OPK-03,OPB-08,OPJ-05,OPP-20,OPT-20,OPZ-09,OPS-12,OPB-14)which contain ten base to amplify polymorphic DNA of seventy-two E^Fs (wheat/ E.rectisetus// wheat) which contain sixteen lines and thirty-six BC3F2(wheat/E.rectisetus//wheat//wheat). E.rectisetus and wheat is served as parental lines to develop for RAPD marker, and to establish the optimum research condition and optimum system of reaction. Lately, the study demonstrates that 84 out of 108 materias got the RAPD marker (the fragment DNA of E.rectisetus which does not contain in wheat ) combination with the result of field agronomy morphology we can reason that there is probably nine type alien addition lines. ? mono-body out of 1045. 3 mono-body out of 1034. 2mono-body out of 1027 and 1 mono-body out of 1035 got the OPE-09750bp marker; (2)5 mono-body out of 1057^ E9721-1-2, E9721-2-2^ E9722-1-K E9716-2-K 1-52-yi-l-l, got the OPN-12i20obP marker; (3)9 mono-body out of 1040 got the OPS-12650bp marker; ? mono-body out of 1048, 4 mono-body out of 1049 and 1 mono-body out of 1055 got the OPE-09750bP and OPZ-09650bP marker; ?055-4-2-1 got the OPZ-0965obP marker; ?059-5-2-1 got the OPT-2050obP marker; ?E9710-l-l,1035-y3-3-l got OPK-036oobP marker; ?1026-1-1-1 got OPB-149oobP marker; î €9709-1-1 got OPZ-0950obP marker.Some of the 9 types have been identified as alien addition lines through cytology techniques and in situ hybridization of nuclear acids. Therefore, the combination of in situ pybridization of nuclear acids with RAPD is a fast and effective method of appraising alien addition lines.
