In the research the representative plant virus TMV(Tobacco mosaic virus) and animal virus Enterovirus and Rotavirus were chosen as detection object. And the viruses in Fuzhou segment of Mm River were detected. After concentrating water sample by chemical flocculation, we detected TMV in concentrated water sample by RT-PCR (reverse transcription-polymerase chain reaction) and traditional biological method. The target band has been amplified from both the concentrated water sample and the tobacco (K3 26) leaf that was inoculated concentrated water sample. The RT-PCR result and the result of inoculation, watching by electron microscope and ELISA (enzyme-linked immunosorbent assay) made it clearly that there were infectious TMV in the water environment. We endeavored to detect animal viruses in water primarily by RT-PCR. We used the primer for enterovirus and for rotavirus respectively and the bands whose sizes were nearly equal to what we expected were amplified. In order to ascertain if there were the two animal viruses in water environment, more experiments were needed. In our research we made an effort on concentrating water sample, extracting nucleic acid from plant tissue and water sample and optimizing the reaction conditions of PCR. Our work was the first time to detect virus in Mm River, and it was the base to improve the detecting method and extending the detecting scope in the field of water virus research.
