Primary shrimp cell cultures were developed from muscle, hepatopand and hemocytes of Macro brachium nipponense in medium 199 supplemented with 20% fetal bovine serum(FBS), 5.2g/L NaCI, I .43g/LCaCI2, 0.05g/LMgC12, 100IU/mL penicillin, 100 ii g/mL streptomycin. Basal medium supplemented with 1 g/L glucose could enhance growth and cells performance. Muscle cells were passaged up to three times. It was found that better results for cell culture would be obtained more easily with the juvenile shrimp caught in spring than the adults in summer or autumn; and shrimps gotten within 12 hours after ecdysis could grow much better than the intermoult shrimps. The osmolality and pH of the different media were tested. The results showed that the optimum conditions for cell culture in vitro were obtained in medium 199 with an osmolality of approximately 57ommol/kg. The favorable pH range of the muscle cells was pH 7.2-8.0, with an optimum of pH7.6. In another way, basal medium supplemented with Zn260 tt g/L could enhance the growth of the muscle cells, The highest concentrations of total phosphorus and the largest RNA/DNA values were found in the groups of Zn2+ 60 ii g/L.
