Applicant for Master Degree: Mao fengxian Specialized Subject: Genetics ,Breeding and Production of Animal Tutor: Zhao youzhang (Gansu Agricutural university 730070) ABSTRACT The experiment was designed to compare the effects of five diluents widely used home and abroad freezing Boer semen. In consequence, the best one is A,which includes Tris 4.361g, glucose 0.654g, citrate 2.399g,yold I 8ml,glycerine 6m1,distilled water lOOml,penicilin and streptomecine each 100,000 units. The researcher compared the effects of rates of dilution, times of cooling, sizes of pellet and thawing temperatures all including three levels on freezing semen. As a result, the ideal pattern means diluting in 1: 2, cooling in 4h, pelleting in 0.2m1 and thawing with VB12 on 37扖. Then I added different concentrions of saccharose, lactose, milk power, raffinose, glutamate or compound vitamin B to diluent A and froze semen. The two best groups are saccharosel and glutamate3. After Comparing the effects of diluents including different concentrations saccharose, glutamate and citrate ,I found the best diluent includes saccharosel, glutamate3 and citric acidl which comprises Tris 4.361g, glucose 0.654g, saccharose 1.6g, citric acid l.972g, glutamate 0.04g, yolk 18m1, glycerine 6ml, distilled water penicillin and streptomecine each 100,000 units. At last a mount of pellet semens were made in the manner. Not returning rate in an oestrus was 65.77% (146/222). It means that the best composition of diluent and freezing craft can be used in practice and an ideal outcoming will come.
