Effect Of Eucommia Ulmoides Ellis Injection On Mastitis In Mice

Mastitis is one of the serious diseases that causes huge financial losses in the aquaculture. We investigate the effect of Eucommia injection that extract from Eucommia ulmoides Oliver against mastitis in mice and expect to provide theoretical foundation or experiment evidence for the prevention and treatment of the mastitis.To address this study, here, we challenged the mouse with the LPS at the concentration of 0.2mg/mL 50 μl by perfusion in galactography. The mice were divided into 6 groups of A,B,C,D,E,F respectively. Among them, A as control group, B as stimulation group, and C,D,E,F as administration group,in test group, the mouse was recevied the volume of Eucommia injection at 0.4 mL, 0.6 mL, 0.8 mL, 1.0 m L respectively. After 12 h of treatment, mammary was dissected and observed the pathological changes via hematoxylin-eosin staining with microscope.Additionally, cytokines including TNF-α,IL-6, IL-1β collected from in infected mice were assessed by ELISA. In another assay, mRNA expressions of ICAM-1, PECAM-1 were detected using quantitative real-time RT-PCR.Results :(1) LPS-induced mouse model could be estabilished successfully by perfusion in the4 th mammary galactography at a concentration of 0.2 mg/mL 50 μL;(2) the results of hematoxylin-eosin staining suggested that Eucommia injection could extenuate the extent of injury, the test groups were observed tha remission of PMN cellular infiltration obviously,furthermore,among all of the treated groups, D and E groups exhibited the effect significantly;(3) Eucommia injection treatment could decease the levels of TNF-α, IL-6, IL-1βexpression in infected mammary tissue, E group demonstrated the best effect(p<0.01);(4)in the LPS-induced mouse model,,the mRNA expression of ICAM-1showed a signinficant increase(p<0.05),while in contrast to the results of treatment,especially E group exhibited clear inhibition comparing with B(p<0.05) and no significant impact.Conclusion: Eucommia injection can alivate pathological injury in breast and significantly decrease the levels of the pro-inflammation cytokines such as TNF-α、IL-6、IL-1β. In addition Eucommia injection also mitigates the stimulation of PMN by cytokines and the migration of PMN to mammary tissue due to the inhibitory of the expression levels in mRNA of ICAM-1,hence,it can extenuate injury in mammary tissue.
