Muscle growth and development are regulated by protein congding genes, and miRNA take part in this process as post-transcriptional factors. To determine the expression patterns of protein-coding genes and miRNAs related to muscle growth and development, after assessing the stablity of six house-keeping gens, we measured the expression level of 25 protein-coding and 16 miRNA genes of pig in skeletal and cardiac muscle at five developmental stages by qRT-PCR, then ascertain their expression patterns by STEM software. The results as follows:1. We evaluated the stability of PPIA, ACTB, TBP, Top2B, YWHAZ and RPL4 by NormFinder and GeNorm software, and selected PPIA, YWHAZ and RPL4 as the house-keeping genes for correcting the expression quantity of mRNAs.2. After STEM clustering, we found in CM, MSTN, NFκB, Smad2, Smad3, S6K1, PDK1, IGF1R and Aktl classified into a same cluster, reached extremely remarkable level (P< 0.001); MURFl, Follistatin, MAPK clustered into one group, reached significant level (P= 0.040). In LDM, MTOR, FOXO1, MRF4, IGF1, MURF1, Smad3, MYHC, SRF, MEF2A, Aktl and IRS1 classified into a same cluster, reached extremely remarkable level (P< 0.001); Smad2, MYF4, MYOD, Follistatin, MYF5 and IGF1R classified into a same cluster, reached extremely remarkable level (P= 0.009). In PMM, NFκB, FOXO1, MRF4, IGF1, MURF1, PDK1, GSK3 classified into a same cluster, reached extremely remarkable leve 1(P= 0.002); Smad 2, MYF4, Follistatin, MYF5, PI3K clustered into one group, reached significant level(P= 0.02).3. STEM cluster reslut of miRNAs:in CM, miR-1, miR-486, miR-221, miR-195, miR-320 and miR-24 clustered into one group, reached extremely remarkable level(P< 0.001); miR-214, miR-181c, miR-181a, miR-499 clustered into one group, reached significant level (P= 0.040). In LDM, miR-1, miR-133a-5p, miR-214, miR-221-3p, miR-499-5p and miR-320 classified into a same cluster, reached extremely remarkable level (P< 0.001). In PMM, miR-1, miR-208b, miR-206miR-486 and miR-499 clustered into one group, reached extremely remarkable level (P-0.003).4. By significance testing and correlation analysia, we found a significant negative relationships between 57 mRNA-miRNA pairs at the five time pionts, like IGF1-miR-1.These results show that these protein coding genes and miRNAs take part in the regulation of muscle growth and development, and present different expression patterns from each other. Our results provided basic information for further investigation of the regulation mechanism of muscle growth and development. |
