Based on the achievement of the molecule biology and the computational iconography, a study on the PET-based CAD/CAM technology of three-dimensional intensity-modulated leaden mould for radiotherapy theory and methodology which contained the intensity-modulated and the biological morphogenesis radiotherapy were presented innovativeness shooting the cancer cell with great accuracy in this paper. First of all,the data of the cancer surface displaying in the PET which was scanned transmission was extracted, and then the three-dimensional focus was reconstructed and the data file was generated in this paper. Referencing the radiotherapy treatment planning system mostly, the concept and transform processing method of the coordinate system were putted forward. In this paper the triangulation was chose after analyzed three methods of calculating the tumor thickness, such as the curved surface fitting, the conical coordinate systems and the triangulation. The triangulation calculated the tumor thickness by spatial geometry in the center coordinate. And the triangulation was accurate and simple. The intensity of ray was evaluation basing the pencil beam kernel and the spatial geometry analyzed the factors which could influence the intensity of ray. And then the point coordinates of the lead mould surface were achieved. And then the lead mould surface which was displayed in the triangulation was amended,smoothed treatment in NURBS and analyzed the error. The ideal clinic lead mould was made eventually. The method was more precise than the method of using the lead mould or the multi leaf collimator. It was objectivity and individuation. So it has great theoretical significance and application value for improving the radiotherapy precision and instrument.
