In this paper, mycelium was obtained from Cordyceps sinensis by liquid fermentation. Crude polyphenols were from mycelia of Cordyceps sinensis. The process of extraction was optimized by orthogonal. Then the crude polyphenol was separated and purified with macroporous adsorbent resins. The kinds and composition of the polyphenols were determined by HPLC. In addition, three classical antioxidant assessment methods and there antitumor activity were studied. The results are as follows:(1)The optimum conditions of extraction were:temperature of 70℃, ethanol concentration of 70%, solid-liquid ratio of 1:35 (g/mL), extracted 60 minutes, two times. Under this condition, the rate of polyphenol extraction was 1.461%.(2)Purification process of macroporous resin for polyphenol was studied. AB-8 macroporous resin showed the best capacity among four kinds of macroporous adsorption resins. The results showed that the optimal process conditions were: sample concentration 2mg/ml, natural pH, flow rate of the sample lmL/min, eluant type 60%ethanol, eluant flow rate 1mL/min. Under the above condition,purity was up to 13.520%from 1.461%.(3)The antioxidant activity of the crude and purified polyphenols were compared by radical scavenging rate and IC50 value with three antioxidant system. The results showed that crude polyphenol and polyphenol purified had high antioxidant and scavenging free radicals activities..The scavenging effects on radical scavenging order is:polyphenol purified>Vc>crude polyphenol.(4)There are differences in the content contained polyphenols of Cordyceps sinensis mycelium. In 19 kinds of polyphenols,5-sulfosalicylic acid, catechin and cinnamon were detected. In which 5-sulfosalicylic acid shows the highest content, respectively 1129.22,2127.99mg/100g.(5)Crude polyphenols and polyphenols purified showed a certain degree of inhibition on Caco-2 colon cancer cells, HepG-2 hepatoma cells and MCF-7 breast cancer cells, in which purified polyphenolic had the best inhibiting effect on HepG-2 hepatoma cells and crude polyphenols showed the best inhibitory effect on MCF-7 breast cancer cells. When the concentration of polyphenols purified was 50μg/mL, the inhibition rate of HepG-2 cell was 83.71%; the inhibition rate of Caco-2 colon cancer cells was 60.71%; the inhibition rate of MCF-7 breast cancer cell was 58.61% The reduction of the number of cancer cells may be affected by Cordycepts Polyphenol antioxidant and may also be affected by the type and content of the polyphenols.
