Basing on the domestic and international research results, its have abundant bioactive functionals like Anticancer Effect antioxidant, antibacterial, cholesterol lowering effect, improve cardio-cerebrovascular cycle, sleep improvement and so on.The domestic and international have further and Systemic study of flavonoids, but the domestic research in limonoids aspect are less. On existing reports, most papers are used Organic Solvent extract limonoids form citrus seed, while the method have disadvantage because of material source not abundant and fall shout of industry produce. This experiment intends to use representational juicing variety orange peel as the material and separate researched acidity and basicity water of combined extraction process Limonoids and flavonoids. On the basis of single-factor test, the mathematical model was established the best parameters through Orthogonal regression of ternary quadratic and response surface methodology. Afterwards preliminary purification of limonoids and flavonoids using as extraction, concentrate, the adsorption separation of the macroporous resin to treatment on extraction solution of orange peel.The acidty and basicitywater use as extraction solution because of enhanced leaching rate, economize, simple equipment, simplify processing, little environmental pollution and combinet extraction process enhanced extraction rate, lower energy, meet the requirement.The main contents and test results are as follows:(1) The first Process is Acid Water Extraction of limonoids, the second process is basicity water extraction flavonoids, after on the comparison the yield rate of acidity and basicity water extraction Limonoids and flavonoids, and comprehensive consideration of chemical properties. Then the separation and purification of united twice extraction solution. (2) This study used the pure water pH regulated by H2SO4 which is the solvent. The limonin was extracted from citrus peels by heat leaching method, and the detection of content of limonin used high performance liquid chromatography. On the basis of single-factor test, the mathematical model was established on the heat leaching extract time, temperature and solid-liquid ratio through Orthogonal regression of ternary quadratic According to this optimum process model and test result analysis, the extraction rate of limonin is 0.117mg/g in the condition of heat leaching extract time:2.6h,temperature:61.2℃,ratio of material to water 1:19.2(g/mL)and the extract solvent pH 4.(3) In this study, the pure water pH was regulated by sodium hydroxide solution.The extract solvent is pH10 basicity of water and flavonoids were extracted from citrus peels by heat leaching method. On the basis of single-factor test, the mathematical model was established on the heat leaching extract time, temperature and solid-liquid ratio through Orthogonal regression of ternary quadratic and response surface methodology. According to this optimum process model, the extraction rate of flavonoids is 0.115% in the condition of heat leaching extract time:2.2h, temperature:64.6℃and ratio of material to water 1:12.(4) The dichlorethane use as extraction adding the united solution, while base on ratio of the dichlorethane to extraction solution 1:1 (ml/ml), extraction time:30min. Organic phase contains limonoids, water phase contains flavonoids.Obtained the crude product of limonoids from the organic phase after volatile the dichlorethane. Adsorption separation of flavonoids is using the macroporous resin in water phase.(5) The adsorption separation property of flavonoids from citrus peels were studied with the macroporous resin. Seven types of macroporous resin were selected to compare their performances in absorbing and desorbing flavonoids and limonoids. The optional macroporous resin was decided and its dynamic absorbing and desorbing behavior were studied. The examination result showed that the macroporous resin purification of limonoids is not different than the extraction process at purity and apart of limonoids lost. This results showed that the macroporous resin can not use in refining limonoids. The examination purification of flavonoids results showed that HPD100 type macroporous resin possess higher absorption and desorption capacity.The absoroption rate of HPD100 resin was 83.5%, the utioeln rate was 92.2%. The optimum technological parameters of the purify of flavonoids by HPD100 are as follows:absorption time 2 hours, desorption time 2 hours,40% alcohol as the desorption solution and rate of absorption content flavonoids to the desorption solution:1:500(mg/ml). |
