The photophysical and photochemical induced by two-photon absorption possess distinguish the especial characteristies of a single photon,which make the two-photon absorption (TAP) materials have a potential application in many flelds. There are certain advantages in the processes of two-photon prossess: absorbing longer wavelength but emitting shorter wavelength with high spatial selectivity and high depth of penetration. The two-photon fluorescence microscope which result from this has made important contributions for the life science research. Due to femtosecond laser with IR output have been used as the excitation, TPM possesses not only high penetration depth of penetration but also without UV-visible photodamage to tissue. However, the lack of two-photon fluorescent probe had limited the development of the TPM imaging. So in a word, it is very important to design and synthesis two-photon fluorescent probe with large two-photon absorption cross sections and good biocom patibility.Five two-photon fluorescent DNA probe with fine two-photon properties and excellent biocompatibility are designd and synthesized in this paper. A series of optical characterization are carried out for them, and study the linear and nonlinear optical properties after the interactions with ctDNA.The main contents of this paper is as follows:1,Five two-photon fluorescent DNA probe of Phenanthroline derivatives with fine two-photon properties designd and synthesized in this paper. Heck and wittig reaction are the main use in the Synthesis of the probe. 1HNMR, IR are utilized for characterization all final compounds.2,UV absorption and fluorescence spectra were measured by UV spectroscopy and fluore scence spectroscopy. Two-photon absorption and polymerization measured with a femto second laser. At the same time, using the reference method of the absorption cross section. Different solvents on the UV spectrum of synthetic materials, single-photon fluorescence, two-photon fluorescence spectra and quantum yield and absorption cross section have been investigated and the solvatochromism has been studied. The relationships between structure and property are discussed in this paper. pH value of the probe molecule 1,2,3,4-term impact is more obvious, but for the salt of the probe molecules are 5 little effect.3,UV absorption spectra, single-photon fluorescence spectra and two-photon fluorescence spectrum have been detailed study between probe molecules and ctDNA. By increasing the concentration of ctDNA, discuss their spectra of linear and nonlinear changes.
