The thesis described the basic principles and the applications of chemluminescence (CL) in biological and pharmaceutical analysis. The main content consisted of two parts:Part I:ReviewIn the first part, based on the fundamental theory of CL and combined with my research work, the applications of various CL systems for biological and pharmaceutical analysis were simply reviewed by using 183 references.Part II:Research reportsLuminol, as one of the most important CL compounds, had been extensively studied. Based on the CL reaction of luminol-hydrogen peroxide and luminol-dissolved oxygen in alkaline medium combined with flow injection, several new methods have been established.(1) Cytochrome c (Cyt c) plays a key role in the biological respiratory chain, whose function is to receive electrons from Cyt c reductase and deliver them to Cyt c oxidase. In this work, it was first found that Cyt c could significantly catalyze the CL reaction from luminol-hydrogen peroxide, and a sensitive and simple procedure was proposed for the determination of Cyt c in a flow injection system. The increment of CL intensity was correlated with Cyt c concentration in the range of 5 to 700 ng mL-1.(2) Hesperetin is one of the most abundant flavonoids. It has multiple biological and pharmacological activities, including antioxidant properties, antiplatelet activity, anti-tumor, etc. It was first found that hesperetin could greatly inhibit the CL intensity generated from the reaction between luminol and dissolved oxygen in alkaline medium. The sensitive FI-CL method was applied successfully to determine of hesperetin in pericarpium citri reticulatae water, human urine and serum samples.(3) Phenobarbital, is a long-acting sedative and anticonvulsant. A simple flow injection CL method with synergistic enhancement has been investigated for the rapid and sensitive determination of phenobarbital.The enhanced CL intensity was linear with the concentration of phenobarbital over the range from 0.05 to10 ng mL-1 with a detection limit of 0.02 ng mL-1. The proposed method was applied successfully in the determination of phenobarbital in human serum and urine samples. The recovery for the phenobarbital samples was from 93.9% to 115.9%.
